Abstract

The aims of this study were to identify and measure the prevalence of alimentary tract worm in stray and domestic cats in the campus area of Semarang State University (Universitas Negeri Semarang/Unnes). As many as 30 cat feces samples: 10 stray cats samples (SC), 10 limited range domestic pet cats (LDC), and 10 indoor domestic pet cats (IDC) were used in this study. Examination of the feces samples was done by using the flotation method to identify the eggs of worms. Based on feces examination, eggs of four worms species i.e. Ancylostoma caninum, Clonorchis sinensis, Dipylidium caninum and Toxocara cati were found. Based the prevalence of worm eggs: A. caninum was found infected 4 out of 30 samples (13.3%), C. sinensis was found infected 2 out of 30 samples (6.6%), D. caninum was found infected 2 out of 30 samples (6.6%), and T. cati was found infected 10 out of 30 samples (33.3%).

Abstract

The aim of this study was to measure antibacterial activity of soursop leaf extract (Annona muricata L.) on bacterial growth inhibition of Pseudomonas aeruginosa by diffuse disc method. The result of the inhibition zone was analyzed using Analysis of Variance and followed using Duncan test, to show highly significantly different (p<0.01). The inhibition number of negative control (C-) was 0.00e±0.00 and the positive control (C+) was 31.31a±0.57. The result of concentration 1 (T1) was 14.49b±4.96; concentration 2 (T2) 8.98c±1.29; concentration 3 (T3) 5.84cd±1.85; concentration 4 (T4) 5.56cd±1.58; concentration 5 (T5) 2.85de±0.28; concentration 6 (T6) 2.98de±0.53; concentration 7 (T7) 2.82de±1.59; concentration 8 (T8) 2.41de±1.10; concentration 9 (T9) 2.20de±0.34; concentration 10 (T10) 1.10e±0.19; and concentration 11 (T11) 0.00e±0.00. The increase of soursop leaf extract concentration showed high inhibition diameter of bacterial growth. It concluded that soursop leaf extract have an antibacterial activity to inhibit P. aeruginosa growth with Minimal Inhibitory Concentration (MIC) 125 ppm.

Abstract

Sperm quality have been associated with age for many years. This study aims to determine the inffluence of age to the volume and motility of Landrace boar’s sperm at Baturiti Artificial Insemination (AI) center. A Total of 300 ejaculates were used in this study. An ejaculate origin from five boar are a collection of five month. A complete randomized design (CRD) was used with two different boar of block ages, block A (2-4 year) and block B (6-8 year). Three or four glass slides were prepared for each boar sample; a drop of semen was placed on each glass slides. This sample was examined under the light microscope on five view field in each glass slides. The result of the research show that the Landrace boars in block A have semen volume average 273.60 mL and sperm motility 73.86%, while in block B the average semen volume was 107.66 mL and sperm motility 62.92%. It can be concluded that Landrace boars in block A had higher volume and sperm motility compared to Landrace boars in block B.

Abstract

Toxocariasis is one of zoonosis diseases that caused by Toxocara spp. that is Toxocara canis. Toxocara canis has several stages until it can infect animals and humans, namely the egg stage, larvae first stage (L1), larvae second stage (L2), larvae third stage (L3) to adult worms. Studies about the L2 and L2 tissue of T. canis found in paratenic hosts using Scanning Electron Microscopy (SEM) have not been widely performed. Some of the causes include L2 being not easily to found and identified, so research rarely raises the ultrastructural morphology of L2 and L2 tissues. Knowledge about the ultrastructural morphology of L2 and L2 tissue of T. canis worms is very important to determining the diagnosis, especially the etiological diagnosis. The purpose of this study is to detected morphology of L2 and L2 tissues of T. canis using SEM. Samples from this study is faeces of dogs that infected with toxocariasis and the digestive tract of dogs obtained from dog slaughter houses. The sample is an adult worm of T. canis; the female worm is dissected and taken uterus to collect worm eggs. The results of this study on microscopic and optilab examination showed a difference between L2 and L2 tissue that the length of L2 hatched from embryonic eggs was 390 ìm and with a width of 23.4 ìm at the midpoint of the body. Larvae second stage length from the infected somatic tissue is 410 ìm and the width is 22.5 ìm at the midpoint, and then difference in dorsal lip, cuticles, body ring, cervical alae, buccal capsul, tail.

Abstract

Grouper are reef fish which can be cultivated because highly commercial value. There are many different types of bacteria in the digestive tract of grouper, one of them is Staphylococcus. Staphylococcus sp. is a Gram positive, non spora, non motile, and facultative anaerobic. Most of Staphylococcus sp. are pathogenic in human and animals. The purpose of this study was to identify the species Staphylococcus sp. on grouper sold at the Kedonganan fish market, Kuta District, Badung Regency, Bali Province based on 16S rRNA sequence analysis. This study used grouper feces as a samples from 20 fishes. Staphylococcus sp. from feces were cultured in media Blood Agar. Sequence analysis 16S rRNA Staphylococcus sp. was conducted with PCR and sequencing method. This study can identify the bacteria Staphylococcus sciuri and Staphylococcus hominis.

Abstract

Mimusops elengi L. , Averrhoa carambola L. and Curcuma xanthorrihiza Roxb singly were reported to have hypothensif effect. Mechanism underlying decrease of blood pressure was suggested through modification of cardiac activity. The Effect of Mimusops elengi L., combined with Averrhoa carambola L. and Curcuma xanthorrihiza Roxb extract on cardiac activity were studied using ECG on 12 male cats that were grouped to be a control group administered aquadestilata, and treatment group administered extract of 21 mg and 82 mg/2 kg bw orally respectively. The extracts were given 3 h prior to ECG. The results showed a decrease in P wave, QRS complex and the speed of the heart rate after administration of combination of extract . However QT and PR intervals were increased. This showed that the extract can weaken a contraction of the Atria and ventricles, prolonging the onset of the occurrence of atrial contraction towards ventricular contraction, extending the onset of contraction and relaxation of the ventricles and cause a decrease in heart rate. Based the cardiogram, It was concluded that their combination is valuable for treatment of aritmia.

Abstract

The aim of the present study was to determine the optimal incubation time of collagenase on concentration, viability, quality of endometrial cell culture, and the role of estradiol and progesterone on ovine endometrial cells proliferation. Endometrium minced was incubated into collagenase with different duration of time: 1 hour, 3 hours, and 6 hours repectively. Cell concentration and viability were calculated after incubation. The quality of cell culture was observed on 1st, 3rd, and 5th day after seeding. The best incubation result was then continued with the addition of the E2 (100 pg/ mL), P4 (100 ng/mL), E2:P4 (100 pg/mL : 10 ng/mL), and E2:P4 (10 pg/mL : 100 ng/mL) into the culture medium. After nine days, cell culture was harvested by trypsinization. Concentration and cell viability were analyzed using analysis of variance, followed by Duncan test. Quality of endometrial cell culture was analyzed descriptively. Results showed that there was a significant decreased in the concentration and cell viability obtained in each treatment of collagenase incubation time and optimum treatment of endometrial cell culture was found at 3 hours. Experiment on culture of endometrial cell revealed that proliferation rate treated by E2 and P4 was better then control (P<0.05). Futhermore, additional of E2 into the culture medium even E2 alone (100 pg/mL or higher E2 combination with P4 (100 pg/mL : 10 ng/mL) showed better proliferation rate than P4 alone (100 ng/mL) or higher P4 combination (10 pg/mL : 100 ng/mL). In conclusion, suplementation of 100 pg/mL of estradiol (E2) could support better proliferation of ovine endometrial cells in vitro.

Abstract

Staphylococcus aureus is a pathogenic bacterium causing disease in humans and animals. In broilers it cause septicemia, tendosinovitis, dermatitis, endocarditis, wound infections and arthritis and bumblefoot. The ability of Staphylococcus aureus to cause disease depends on the virulence factors they bear. The purpose of this research is to investigate the distribution of pigment production type and the existence of genes of polysaccharide capsule phenotype and genotype as determinant factor of virulence of bacteria on 15 isolates of Staphylococcus aureus from broiler. Pigment production test showed that 86.7% of isolates producing yellow pigment and 13.3% isolates produce orange pigment. The detection of polysaccharide capsules was phenotypically performed with hydrophobicity test with serum soft agar medium (SSA) showed 53.3% isolate grow compact and 46,7% isolate grown difuse and hydrophobicity test by salt aggregation test method (SAT) showed 66,7% hydrophil and 33.3% are hydrophobic. Genotype detection of polysaccharide capsule genes by polymerase chain reaction (PCR) showed 66.7% detected cap5 (amplicon 361bp) and 33.3% detected cap8 (173bp ampliole). The type of pigment production and the presence of polysaccharide capsules are some of the virulent factors in Staphylococcus aureus.

Abstract

Newcastle disease is a pathogenic viral disease in poultry which is infectious and can cause massive economic losses. The disease is still endemic in Indonesia. To understand the pathogenesis and the distribution pattern of the virus in the tissues, pathological observation was evaluated using newly virulent isolate Newcastle disease virus (NDV) that was inoculated in embryonated chicken eggs. As many as seven embryonic chicken eggs aged 11 days and specific antibody negative against Newcastle disease, divided into two categories: inoculated with phosphate buffer saline and inoculated with isolates. Then the allantois fluid was tested using hemagglutination assay and hemagglutination inhibition tests to prove the infection serologically. The hearts, lungs, livers and small intestines of the inoculated products were collected and followed with the process of histopathological preparation using Hematoxylin and Eosin (HE) stain. The pathological analysis showed that all organs had necrosis, hemorrhages, inflammation, and congestion. Congestion and hemorrhages in the hearts only occurred at 60% of the samples. However, necrosis, hemorrhages, and inflammation that were observed in liver occurred at 60%, 40% and 60% of the samples, respectively. Furthermore, the hearts were edema, thinner in the heart muscle fibers; while in the lungs, proliferation of pneumocyte type II was founded. Our finding provided valuable insight into the pathology of a virulent isolate of NDV which is dominated by blood circulation disorders with necrosis and inflammation in the chicken’s embryos and have important implication for the future studies.

Abstract

Bone is a tissue that has a density of extracellular matrix structures and composed by organic and inorganic components. Decalcification is a stage that plays an important role in bone histology using various types of solutions. The sample used in this study was lateral condyle from tibia of three garut sheeps (Ovis aries) which had been fixed with 10% Neutral Buffered Formalin (NBF) for 24 hours. Sample were cut into pieces ranging from 1 cm x 1 cm x 1 cm in size, the decalcification using three solutions; 10% nitric acid, 10% EDTA (pH 7.4) and 10% EDTA (pH 7.4) + TBD-1®. The aim of this study was to evaluate the effectiveness of three solutions for the decalcification process of lateral tibial condyle of garut sheep. Observation parameter in this study includes: duration of decalcification, sectioning process of ribboning formation, structural integrity and absorption of hematoxylin-eosin (HE) staining. The results shows that 10% EDTA (pH 7.4) solution provides a long duration of decalcification which is ease sectioning process ribboning, the best structural integrity of lateral tibial condyle. In hematoxylin-eosin (HE) staining shows that 10% nitric acid solution does not absorb the optimum color, opposite in 10% EDTA and 10% EDTA + TBD-1® solution, the color intensity between hematoxylin and eosin in the tissue shows the best results. Based on these results, it can be concluded that the 10% EDTA (pH 7.4) is the best decalcification solution for lateral condyle from tibial of garut sheeps.
